We are interested in sourcing a glucose-responsive aptamer with a length of approximately 45–60 nucleotides, incorporating boronic acid-modified thymidines at 2-4 specific positions in the loop region. We require 5′-DBCO and a 3′-inverted dT cap for end modifications, along with nuclease protection via 2′-OMe caps on the terminal bases and phosphorothioate bonds at the ends. Additionally, we're looking for a lock strand that's around 12–15 nucleotides, complementary to a segment of the aptamer. It should feature a 5′-PEG(2 kDa)-maleimide for thiol-modified insulin coupling, with an optional 3′-inverted dT. Nuclease protection should include 2′-OMe bases and/or phosphorothioate bonds at the ends.
